The mechanism of action of JAK inhibitors adapted from Shawky, Ahmed M, et al. Janus kinase (JAK) inhibitors block the kinase domain of cytoplasmic non-receptor tyrosine kinases (JAKs), preventing the phosphorylation of STAT transcription factors. By disrupting the JAK-STAT pathway, they reduce cytokine-mediated signal transduction that drives inflammatory and autoimmune responses. This inhibition interferes with the pathogenic immune activation pathway, which promotes inflammation, immune response, and cancer.
